10 X PBS Protocol

 

1.     Take about 500 ml 3A water in a bottle and put a stirrer in it.

2.     Place the bottle on a Magnetic stirrer and turn on the machine.

3.     Add 40 gm NaCl (Tissue culture tested).

4.     Add 1 gm KCl (Tissue culture tested).

5.     Add 7.2 gm Na2HPO4 (Tissue culture tested).

6.     Add 1.2 gm KH2PO4 (Tissue culture tested).

7.     Continue stirring up to dissolve everything

8.     Prepare PH meter for reading PH and then submerge the probe in 10X PBS.

9.     Read the PH and adjust it to 7.4 by adding 10N NaOH and con. HCl. Wear gloves before using them.

10.   After adjusting PH pour the liquid in a graduated flask (500 ml) and adjust the volume to 500 ml finally by adding 3A water.

11.  Pour it again in the bottle and autoclave it.

12.  Preserve the 10X PBS in room temparature
